ABSTRACT: The Echo series of acoustic liquid handlers is widely accepted as a gold standard in compound transfer and high throughput screening (HTS) applications. In this tutorial, we will present two studies to show how Echo Liquid Handlers enable workflows beyond HTS to streamline genomic applications. Identifying acquired mutations not present in normal patient tissue, that exist in tumor derived material, is important for diagnostic testing for cancer, and can ultimately inform targeted therapy. In the first study, we demonstrate a highly efficient, precise and accurate, low-cost method to detect PIK3CA somatic mutations. The miniaturization of the PIK3CA mutational analysis assay results in significantly less input DNA required, allowing conservation of precious and limited samples. The Nextera method to create libraries for Illumina sequencing has steps that limit broad utility due to cost and throughput. In the second study, we show how the Echo 525 liquid handler eliminates these barriers and enables a robust workflow that supports both rapid and high-throughput sample preparation while significantly reducing reagent costs. |

ABSTRACT: Learn more about AstraZeneca’s collaboration with Labcyte, Brooks Life Science Systems, and Titian Software to develop the automated storage systems, liquid handlers and software required for the implementation of an Labcyte® Echo® Qualified Microtube. At this event, attendees will hear about AstraZeneca’s mission to transform their sample management and screening programs with a broader implementation of acoustic liquid handling. As AstraZeneca increases the diversity of its screening library, the ability to dispense nanoliter volumes of sample directly from low-volume storage tubes will enable screening of rare or difficult to synthesize samples and reduce storage costs. Additionally the rapid cherry picking and plating of samples will enable a more efficient creation of target-specific libraries for iterative screening programs. Through their collaboration with leading storage, acoustic liquid handling and software providers, AstraZeneca is driving the commercialization of a fully automated system to better address the future of sample management. |
